I know I could just paint the triangle with the same colour behind, but I will be exporting with an Alpha, so I would prefer to have the triangle transparent.
I tried both Clip and Mask, but either these are not the right tool for this, or I just don't know how to use it.

You can use clipping to do it, and I could tell you how. But I would use Difference.
-- Select the letter.
-- Path menu > Break Apart
-- Deselect
-- Select the triangle path
-- Object menu > Raise to Top (or this button on Selection tool control bar :mtt: )
-- Select both paths
-- Path menu > Difference
