Hi All

I have used 0.49 on Window XP.

Here I am drawing in one Square Filling with Stripes 1:1 and Save File as .plt.
When i again open the same file ,it shows only boundary( without Strips) on screen.

Let me know how can i solve this issue.

PLT doesn't support fills, let alone pattern fills, and Inkscape isn't yet able to outline pattern fill of an object. So you will have to find another way to do what you wanted to do.

Note -- I moved this topic to the Cutters/Plotters forum, but upon re-reading it, it's not clear that you're planning to cut or plot. So if this is entirely the wrong place, let me know and I'll move it instead to the Help forum.
